contact At COOLLED, we are pleased that the product has been so positively received by many experienced fluorescence microscopists who have recognised the benefits of LED technology, particularly the new family of high-power LEDs in the precisExciteŽ product. During the development of this product, Southampton University was used as its Beta test site facility. "precisExcite units have replaced mercury bulb fluorescence systems in our microscopy facility with great success. A straightforward plug and play design, an easy to use control pad, adjustable intensity and fluorescence output stability has made it popular with our large user group. It is easily bright enough for all our current projects, including live cell samples, fixed tissue sections, and plant material; even penetrating deep into drosophila larvae preparations. As facility manager, I am happy that we no longer have to deal with the safety issues surrounding mercury bulbs, and even happier that I will never have to carry out bulb alignment again. We estimate that with each of the three wavelengths having 10,000 hours life expectancy, that we won't be changing our fluorescent sources for the next 15 to 25 years!" Dr. Matt Cuttle, During the development of this product, we asked the Laboratory for Molecular Biology at Addenbrookes, Cambridge to comment on the product. Light emitting diode units are now available, with emission peaks at 400nm (DAPI excitation), 460nm (FITC) and 520nm (TRITC). These also are connected via a liquid light guide. The diodes can be turned on and off rapidly at will, or under computer control, last for more than 50,000 hours and are much more stable in intensity than arc lamps. [With the precisExcite product,] I found no variation in intensity over 9 hours of recording. If you are using your mercury lamp as an adjunct to a confocal laser scanning microscope, for time-lapse recording or for any other purpose where the highest possible intensity is not necessary, it would be worth switching to the LED.
